Job Summary The Gene and Cell Therapy Institute (GCTI) is a newly created system-wide Institute under Mass General Brigham (MGB). The Institute will foster a world-class gene and cell therapy program that will leverage ongoing, groundbreaking scientific research efforts across the MGB system. The mission of GCTI is to bring together the community of MGB investigators which can utilize gene and cell therapy platforms to achieve their scientific and clinical goals and improve the health of patients. The Institute will differentiate itself by harnessing the exceptional strengths of clinician scientists, the large and unique patient population, and major discoveries throughout the hospital system to propel MGB as the leader in Gene & Cell therapies.The Gene Editing Core (GEC) at the GCTI, Mass General Brigham, is seeking a highly motivated Research Technician II to support genome editing, CRISPR-based perturbation screening, iPSC reprogramming, and associated molecular and cellular workflows. The successful candidate will perform mammalian cell culture, molecular biology, genome editing, and quality-control assays in support of research projects from investigators across MGB and external collaborators. This position provides an opportunity to gain hands-on experience with advanced genome-editing technologies in a collaborative core facility environment. The successful candidate will have experience working in a research laboratory environment and proficiency in standard molecular and cell biology techniques, including mammalian cell culture, DNA/RNA isolation, PCR, gel electrophoresis, and basic molecular cloning. Experience with CRISPR-based genome editing, iPSC culture, lentiviral vector production, and/or next-generation sequencing workflows is preferred. The candidate should be detail-oriented, well organized, able to follow and document standardized protocols, and comfortable managing multiple projects and samples simultaneously. The individual should also be highly motivated to learn new gene-editing technologies and work collaboratively in a fast-paced core facility environment supporting multiple research groups. Essential Functions -Support genome editing, CRISPR-based perturbation screening, iPSC reprogramming, and lentiviral vector production workflows. -Maintain and culture mammalian cell lines, including human iPSCs and primary cells, using aseptic technique. -Perform routine molecular biology procedures, including DNA/RNA extraction, PCR, qPCR, gel electrophoresis, and plasmid preparation. -Support CRISPR-based genome-editing workflows, including reagent preparation, transfection/nucleofection, single-cell clone isolation and expansion, and genotyping of edited clones. -Support next-generation sequencing (NGS) workflows, including amplicon/library preparation, sample quality control, and preparation of samples for sequencing. -Perform cell characterization and quality-control assays, including immunofluorescence staining, microscopy, and other established assays. -Maintain accurate experimental records, sample inventories, and project documentation according to established core SOPs. -Perform routine data collection, organization, and basic analysis; communicate experimental results to core staff. -Assist with laboratory inventory, reagent preparation, equipment maintenance, and general core operations. -Follow established SOPs, biosafety requirements, and laboratory quality practices. -Assist with troubleshooting and optimization of established gene-editing and molecular biology workflows under appropriate supervision. -Participate in team meetings, training activities, and continuous workflow improvement.
