This11-week summer internship is designed forseniorundergraduate, Master's, or PhD studentsseeking exposure to a professional, corporate environment. The internship is project-based, with each intern assigned one clearly defined project aligned to the hosting department's priorities.

The role emphasizes learning, skill development, and practical application of academic knowledge while contributing to meaningful work.

Project Scope:

The goal of this project is to compare two Neon electroporation systems in terms oftransfection efficiency,cell viabilitypost transfection, andbulk pool titer. The two systems will be evaluatedside by sideunder matched experimental conditions. The project will include the following experimental components:

  1. GFP Transfection Efficiency Assessment
  • Transfect CHO host cells with a GFP plasmid.
  • Measure the percentage ofGFP positivecells using theGUAVA flow cytometer.
  1. TPOR DNA Preparation
  • Linearize theTPORplasmid DNA.
  • Perform purification of the linearized DNA to ensure high-quality material for transfection.
  1. TPOR DNA Transfection and Bulk Pool Development
  • Transfect CHO host cells with the linearized TPOR DNA.
  • Initiate bulk pool development with selection pressure.
  • Monitorcell viabilityandVCDthroughout pool expansion.
  1. Bulk Pool Titer and Product Quality Analysis
  • Continue bulk pool development until material is ready to set up fed-batch.
  • Measuretiterand evaluateproduct quality attributes.

Summary:

By comparingtransfection efficiency,post transfectionviability, andbulk pool titeracross the two electroporation systems, we willdeterminewhether the newNxTElectroporation Systemcan effectively replace the legacy system.
